Performs a range of molecular biology, cell biology, epigenomic, and in vivo laboratory procedures in support of a translational sarcoma research program focused on ATRX-deficient soft tissue sarcomas (Nacev Laboratory, Division of Hematology/Oncology, UPMC Hillman Cancer Center). Core duties include maintaining and manipulating human and murine sarcoma cell lines, performing drug treatment and cell viability assays; immunoblotting; chromatin and epigenomic profiling (CUT&RUN, ATAC-seq); nucleic acid extraction and next-generation sequencing library preparation (RNA-seq, whole-genome bisulfite sequencing); and murine allograft tumor studies conducted under an approved IACUC protocol. The technician also supports general laboratory operations, including supply management, equipment maintenance, and accurate, timely experimental record keeping in accordance with good laboratory practice (GLP).
Performs cell culture and maintenance of human and murine sarcoma cell lines
Conducts drug treatment and dose-response cell viability assays (e.g., ATP-based luminescent viability assays)
Performs molecular and biochemical assays including immunoblotting, PCR, and nucleic acid (DNA/RNA) extraction
Performs chromatin and epigenomic profiling assays such as CUT&RUN and ATAC-seq, and assists with sequencing library preparation for RNA-seq and whole-genome bisulfite sequencing (WGBS)
Assists with murine studies, including allograft tumor inoculation, tumor measurement, drug administration, necropsy, and tissue harvest, under an approved IACUC protocol
Identifies and prepares specimens; maintains detailed, accurate, and timely experimental records, data, and logbooks; organizes and documents results
Ensures compliance with institutional biosafety (BSL-2+), IACUC, and laboratory safety requirements
Minimum requirements: Bachelor's degree in a biological science or related field and a minimum of 1 year of relevant laboratory research experience.
Preferred: Hands-on experience with mammalian cell culture, drug treatment and dose-response testing, and animal handling. Prior experience with chromatin/epigenomic assays (CUT&RUN, ATAC-seq) and murine tumor models is a plus but not required; training will be provided.
*This position will work on Dr. Nacev's newly funded DoD TOPPLE grant. Funding for this position is available for 2 years after which the position will no longer be funded. *
